Expansion Microscopy (ExM)
As part of the DEEP-DV Research Unit’s training program, we had the opportunity to learn the technique Expansion Microscopy (ExM) directly from experts at CSSB in Hamburg. This hands-on course introduced us to a powerful imaging technique that enables super-resolution microscopy without specialized equipment by embedding samples in an expandable gel, physically enlarging the specimen and revealing structures below the usual resolution limit.
Over three days, we carried out the full ExM workflow – from sample preparation and gel embedding to expansion, staining, and imaging with a spinning disc confocal microscope. Through this training, we could visualize subcellular structures such as the microtubule organizing center and HCMV virions (~200 nm), gaining valuable practical experience with a method that we can now apply in our own research.
Beyond the technical skills, the workshop highlighted the strength of the DEEP-DV training environment. The supportive atmosphere, expert guidance, and interaction with fellow PhD students made it easy to engage with a completely new technique and build confidence quickly. Overall, this experience significantly broadened our methodological skillset and demonstrated the value of the high-quality training opportunities provided within DEEP-DV. A big thank you to teachers Yannick Jensen and Timothy Soh, members of the Quantitative virology Research Group led by Jens Bosse, who expanded the knowledge of DEEP-DV.
